Isolation and Phenotypic Characterization of Lytic Bacteriophage with Activity against Pseudomonas aeruginosa

Aims: To perform the isolation and phenotypic characterization of bacteriophage with lytic activity against Pseudomonas aeruginosa. demonstrate that this type viral agent can be isolated from environment used for biocontrol resistant bacterial types, such as aeruginosa.
 Study Design: This study was an experimental study.
 Place Duration Study: The conducted at, Bacteriology Mycology Laboratory in Veterinary Hospital at School Agricultural Sciences, Innovation Business University Passo Fundo (ESAN/UPF) Center Diagnosis Research Animal Health (CDSA/UPF), between April 2022 June 2022.
 Methodology: Samples untreated water were inoculated host bacterium strain aeruginosa ATCC 27853 enriched media After incubation period in, a phage filtrate obtained by centrifugation followed filtration. We verified presence bacteriophages using spot test we carried out its purification method sterile toothpick plate transfer on overlay semi-solid agar. Amplification performed SM buffer elution procedure to produce stock material. Through assays Petri dishes overlay, titration regarding lysis spectrum efficiency infection host.
 Results: managed isolate morphologically characterized approximately 1 mm diameter, high clarity inhibition area, halo well-demarcated edges. bacteriophage, named Phage UPF_PaBP1, demonstrated capacity target bacteria all tested dilutions preparation titre 6.5 x 10⁸ PFU/ml future use.
 Conclusion: showed strong host, finding nourishes our expectations use therapy.
